Item Cardiac changes in spontaneously hypertensive rats : modulation by aerobic exercise.(2023) Teles, Maria Cecília; Portes, Alexandre Martins Oliveira; Coelho, Bianca Iara Campos; Resende, Letícia Teresinha; Isoldi, Mauro CésarSystemic arterial hypertension is a multifactorial clinical condition characterized by high and sustained levels of blood pressure. For a better understanding of the pathophysiology of hypertension, studies are conducted with spontaneously hypertensive animals, which allow the investigation of physiological changes that in most cases cannot be studied in humans. In these animals, myocardial remodeling, increased pro-inflammatory markers, redox imbalance and contractile dysfunctions that lead to changes in cardiac function can be observed. However, it can be inferring that aerobic training improves cardiac function and cardiomyocyte contractility, in addition to controlling inflammation and reducing oxidative stress in cardiac muscle, despite this, the precise mechanisms by which physical exercise improves cardiovascular control are not fully understood. In this review, we provide an overview of the pathophysiological changes that affect the heart of spontaneously hypertensive animals and their modulation by aerobic exercise.Item Effects of an acute swimming session until exhaustion in myocytes isolated from Wistar rats.(2022) Teles, Maria Cecília; Caldas, Lucas Rogério dos Reis; Barros, Luciana Fernandes; Coelho, Bianca Iara Campos; Silva, Franciany de Jesus; Isoldi, Mauro CésarThe role of regular physical exercise is recognized for the prevention, control and treatment of cardiovascular diseases. However, studies show that after strenuous aerobic exercise sessions in healthy individuals can lead to cardiac damage, due to the high degree of stress imposed on myocardial structures. This study verifies the effects of swimming to exhaustion on contraction and relaxation velocities, as well as the velocity of calcium release and reuptake in cardiomyocytes isolated from the left ventricle. Wistar animals aged 16 weeks were submitted to a protocol of swimming until exhaustion with a load of 5% of body weight and later the cardiomyocytes were isolated. It was observed that an acute session of swimming until exhaustion promoted an increase in the velocity of contraction and relaxation and an increase in the velocity of calcium release. Exercise to exhaustion promotes adverse effects on the myocardium, however more studies are needed to explain these effects and demonstrate the molecular mechanisms involved in the process.Item Exposição ao frio e seus efeitos na expressão gênica do coração de camundongos alimentados com high-fat diet.(2023) Coelho, Bianca Iara Campos; Isoldi, Mauro César; Isoldi, Mauro César; Silva, Fernanda Cacilda dos Santos; Teles, Maria CecíliaA melanopsina (OPN4) é um fotorreceptor e termo sensor cuja expressão é observada em vários órgãos e tecidos, sendo o coração altamente expresso. Além disso, a expressão dos canais de potencial receptor transitório (TRP) parece estar associada à expressão de OPN4 na retina e supostamente no coração. Partimos da hipótese que a OPN4, juntamente com os canais TRP no coração, atuam monitorando as variações de temperatura interna através do sangue proveniente dos pulmões, possivelmente promovendo a liberação e expressão de seus principais hormônios, os peptídeos natriuréticos (NPs). Fatores como a temperatura do ambiente e alimentação desiquilibrada, prejudicam a homeostase cardíaca, causando efeitos prejudiciais na sua estrutura e função, potencialmente desencadeando doenças cardiovasculares. Portanto, o objetivo do estudo é avaliar os efeitos da exposição ao frio e a ausência do canal TRPA1 sobre a expressão gênica de Opn4, Trpv1, Trpa1, Trpm8, Nppa e Nppb, em átrios e ventrículos do coração de camundongos alimentados com high-fat diet. Foram utilizados 24 camundongos machos C57BL/6J e Trpa1 KO, divididos em 4 grupos experimentais e expostos a temperaturas de 30°C e 22°C. Os resultados de PCR quantitativo indicaram que a expressão de Opn4 nos átrios, à 30°C foi reduzida no grupo Trpa1 KO, em 22°C houve aumento na expressão do gene no mesmo grupo e, em ventrículos não apresenta diferenças significativas. Quanto aos canais TRP, no gene Trpv1 não encontramos diferenças significativas. Trpa1 átrios não houve diferenças significativas, ventrículos à 22°C indicaram redução na expressão. Trpm8 átrios, à 30°C temos redução da expressão do gene no grupo Trpa1 KO, à 22°C resultou em redução na expressão em camundongos C57BL/6J, enquanto, que no grupo Trpa1 KO, houve aumento da expressão. Já em ventrículos, à 30°C resultou em aumento na expressão de Trpm8 no grupo Trpa1 KO e, em C57BL/6J aumento na expressão à 22°C. Nos NPs, a expressão de Nppa não apresentou diferenças significativas. Nppb em átrios, indicou redução na expressão à 22°C, tanto em C57BL/6J como Trpa1 KO. Já em ventrículos, Nppb não apresentou diferenças significativas. Podemos concluir que a OPN4 e os canais TRP possivelmente possam estar atuando como termo sensores no coração, e as flutuações de temperatura afetam a expressão dos NPs nas câmaras cardíacas.Item The effect of diet on the cardiac circadian clock in mice : a systematic review.(2022) Paula, Ana Beatriz Rezende; Resende, Letícia Teresinha; Jardim, Isabela Alcântara Barretto Araújo; Coelho, Bianca Iara Campos; Miranda, Denise Coutinho de; Portes, Alexandre Martins Oliveira; Teles, Maria Cecília; Castrucci, Ana Maria de Lauro; Isoldi, Mauro CésarCircadian rhythms play important roles in regulating physiological and behavioral processes. These are adjusted by environmental cues, such as diet, which acts by synchronizing or attenuating the circadian rhythms of peripheral clocks, such as the liver, intestine, pancreas, white and brown adipose tissue, lungs, kidneys, as well as the heart. Some studies point to the influence of diet composition, feeding timing, and dietary restriction on metabolic homeostasis and circadian rhythms at various levels. Therefore, this systematic review aimed to discuss studies addressing the effect of diet on the heart clock in animal models and, additionally, the chronodisruption of the clock and its relation to the development of cardiovascular disorders in the last 15 years. A search was conducted in the PubMed, Scopus, and Embase databases. The PRISMA guide was used to construct the article. Nineteen studies met all inclusion and exclusion criteria. In summary, these studies have linked the circadian clock to cardiovascular health and suggested that maintaining a robust circadian system may reduce the risks of cardiometabolic and cardiovascular diseases. The effect of time-of-day-dependent eating on the modulation of circadian rhythms of the cardiac clock and energy homeostasis is notable, among its deleterious effects predominantly in the sleep (light) phase and/or at the end of the active phase.
